The right answer is probably Planes Trains, but I voted Weird Science for the happy memory of it.
I remember hanging out and staying up all night with my girlfriend, and walking to 7-11 at 2 am for some awful food, like nachos and Slurpee's - we also grab a movie - we're tired and bleary eyed, and maybe that helped, because the comedy was probably made for 14-year-old boys (we are in our 20s), but we can't stop laughing, we're tickled over every little silly thing. Just in stitches.
I like Ferris and Planes and others, but that was the most fun I had with a Hughes film. Probably wouldn't be the same if I watched it today, but for the good memory, that's my pick.
