The Joker will appear in a flashback scene in Batman v Superman: Dawn of Justice, and it will show us just how the Joker ends up in prison, and it also will give us some insight into why the Batman we see in Zack Snyder’s film is such a screwed up individual.
Apparently, the flashback scene will feature the Joker beating the hell out of Robin and killing him, which comes from the 1988 Batman comic story arc "A Death In The Family," which focused on the Jason Todd version of Robin.

Doomsday WILL appear as one of the main villians. As for the " Do you bleed" scene in the film that we've seen in the trailer, it's actually a dream sequence.

Agreed, plus in the comic it's actually Green Arrow who "beats" Superman by hitting him with a kryptonite arrow anyway.
Ummm, No. I thought we are beyond this nonsense? If you want to go with that logic it still fails, because after Green Arrow hits him with kryptonite (Batman built and supplied Green Arrow with it) Batman still gives last punches...

The movie's called "Batman vs. Superman" and so far in addition to them, we've got
Doomsday
Wonder Woman
Aquaman
Cyborg
And now possibly:
Joker
Robin
Maybe even Lex Luthor (since he was casually referenced in Man of Steel)
Here's hoping they take a cue from Marvel, and release DVD extras which delve into the backstories of these characters, instead of trying to cram 8-9 superheros/villains along with their backstories into a 2 hour movie.
Or just make it an hour longer; I don't see the need to limit it to a strict 2 hours anyway, it's not like people aren't going to see it (the Dark Knight Rises was 2H 45M long and it was a blockbuster).
